Abstract


This study was carried out to investigate the cytotoxicity of cadmium in cultured rat fibroblast and neuroglial cells. Neutral red(NR) and tetrazolium MTT(MTT) of the colorimetric assay were performed to evaluated the cytotoxicity of cell
organells. And
also the lactate dehydrogense(LDH). Activity was measured for the membrane toxicity. The light microscopic study was carried out to morphological changes of cultured rat fibroblasts and neuroglial cells.
@ES The results as follows :
@EN 1. The NR and MTT absorbance were decreased as dose of cadmium increment both fibroblasts and neuroglial cells, especially remarkable in fibroblast.
2. The LDH(Lactate dehydrogenase activity was increased with the increase of cadmium-dose both fibroblasts and neuroglial cells.
3. Light microscopic studies, the morphological changes were showed in fibroblasts and neuroglial cells according as increment of cadmium concentration. In fibroblasts, the typical pyramidal shape was changed to rounded shape and cell necrosis
was
showed, the neuroglial cells showed necrosis of cellular branches decrement as increment of cadmium concentration, respectively.
The results indicated that cadmium was damaged to cell organelles and membrane both of the cell type, fibroblast was more severe than neuroglial cell at same dose of cadmium. In morphological changes, the shape chanes of fibroblast and neuroglial
cells
occurred with increment of cadmium concentration, especially the cellular branches of neuroglial cells was decreased by dose dependently.
